In the industrial era, the need for enzymes continues to increase, for example, proteases. Protease is an enzyme that can accelerate the hydrolysis of protein into amino acids. One of the protease enzymes in the form of papain which is often used for meat tenderizer is the papaya plant (Carica papaya L.). The objective of this study was to obtain endophytic bacterial isolates of the Carica papaya L. plant and determine their proteolytic activity. Endophytic bacteria were isolated from the root, trunk, petiole and leaves of Carica papaya L. Characterization of endophytic bacteria was carried out by macroscopic and microscopic observations. Proteolytic anchovies were screened using Skim Milk Agar and Nutrient Gelatin media. Then the three bacterial isolates with the highest proteolytic index were counted their proteolytic activity every 4 hours for 48 hours. The isolation results obtained that one endophytic bacterial isolate came from the root, four from the petiole, and fourteen from the leaf, while the endophytic bacteria on the stem could not be isolated. The results of screening for proteolytic bacteria contained eight endophytic bacterial isolates that were only able to hydrolyze casein substrate, one isolate was only able to hydrolyze gelatin substrate, four isolates were able to hydrolyze casein and gelatin substrates, while six isolates were unable to hydrolyze casein and gelatin substrates.
