Carbon sources are generally needed by microbes for their growth and development, carbon sources are also very influential in the production of antimicrobial compounds. Endophytic bacteria found in ciplukan root with code isolate A1 have been shown to inhibit the growth of E. coli and S. aureus bacteria. An optimization way to increase the inhibition of endophytic bacteria is to add carbon sources with different types that can increase the activity of bacteria in growth media and is used with various types of concentrations. The purpose of this study was to determine the effect of carbon sources on antibacterial activity in inhibiting the growth of E. coli and S. aureus bacteria and to determine the optimum concentration to produce inhibitory power. The research methods included rejuvenation of endophytic A1 isolates, manufacture of carbon source medium, inoculation of endophytic bacteria, manufacturing of bacterial cultures, antibacterial activity testing and data analysis. This study uses a Completely Randomized Design using two types of carbon sources and three types of concentrations, namely 1%; 0.5% and 0.1%. Data were analyzed using two-way ANOVA and Duncan's continued test. The results of this study indicate an increase in the growth of endophytic A1 isolates with the addition of carbon sources. The results of activity tests on endophytic A1 isolates against E. coli produced a greater inhibition zone at a concentration of 1% for glucose and a concentration of 1% for fructose while against S. aureus produced a greater inhibition zone at a concentration of 1% for glucose and a concentration of 1% for fructose. The results of the analysis using two-way ANOVA have a significant effect (p <0.05) of the type of concentration used. This study concluded that the addition of carbon sources with various types of concentrations that can increase the antibacterial activity of A1 isolates in inhibiting the growth of test bacteria.
