Purification needs to be done since the impurity components are still obtained during production by fermentation. The aim of this research is to detect 5-ALA compounds after purification by ion exchange techniques, and to differentiate the 5-ALA concentrations in the ion exchange purified eluate. Research methods include subculture of bacterial isolates of strain B10, analysis of bacterial growth and detection of 5-ALA, pH measurement, 5-ALA fermentation, 5-ALA crude production, ion exchange purification, 5-ALA analysis with TLC test and UV-Vis spectrophotometer (553 nm), as well as data analysis. This study used a single factor that distinguishes 5-ALA levels after three times elution by ion exchange techniques using the Paired Sample T-test. The results of the study showed that the TLC test had a pink stain, indicated the 5-ALA contained. The test results with a UV-Vis spectrophotometer showed the highest 5-ALA concentration in the second eluate. The results of the analysis using the Paired Sample T-test, exhibited no significant difference (Sig.>,0.05) from the three eluate results of 5-ALA purification.
