Abstract

Daun mimba (Azadiracta indica A. Juss) merupakan tanaman obat yang banyak diteliti memiliki aktivitas antidiabetes dan antioksidan, namun kajian in silico yang secara spesifik mengevaluasi peran quersetin sebagai senyawa utamanya terhadap beberapa target molekuler kunci diabetes melitus masih terbatas. Oleh karena itu penelitian ini bertujuan untuk mengkaji potensi aktivitas quercetin terhadap Peroxisome Proliferator-Activated Receptors-Gamma (PPAR-đ›Ÿ), Dipeptidyl Peptidase-4 (DPP-4), serta target yang berperan dalam pembentukan Reactive Oxygen Species (ROS) menggunakan pendekatan structure-based drug design melalui metode molecular docking. Hasil simulasi docking menunjukkan quersetin memiliki afinitas yang kuat dan stabil terhadap reseptor PPAR-y dan DPP-4, yang ditunjukkan oleh nilai energi ikatan bebas yang rendah dan pembentukan interaksi molekuler penting berupa ikatan hidrogen dan interaksi hidrofobik pada sisi aktif reseptor. Quersetin juga menunjukkan interaksi yang relevan dengan target protein terkait ROS, yang mengindikasikan potensi aktivitas antioksidan melalui penghambatan jalur pembentukan stress oksidatif. Berdasarkan profil afinitas ikatan dan pola interaksi molekuler tersebut, quersetin pada daun mimba berpotensi memiliki aktivitas antidiabetes melalui mekanisme aktivasi PPAR-y dan inhibisi DPP-4, serta berkontribusi dalam menurunkan stres oksidatif.
