Abstract

Merokok berkontribusi pada peningkatan kadar radikal bebas, yang menyebabkan stres oksidatif dalam tubuh, yang ditandai dengan peningkatan kadar malondialdehida (MDA). Malondialdehida adalah produk akhir peroksidasi lipid yang mencerminkan perubahan DNA, senyawa basa teroksidasi, dan sangat terkait dengan penyakit degeneratif. Penelitian ini bertujuan untuk mengevaluasi potensi Syzygium oleana dalam menurunkan kadar MDA. Penelitian ini meliputi pengujian antioksidan, skrining dan identifikasi fitokimia, serta analisis in silico. Pengujian antioksidan pada teh Syzygium oleana menunjukkan IC₅₀ sebesar 69 ppm, menunjukkan aktivitas antioksidan yang kuat. Analisis fitokimia dalam teh mengungkapkan hasil positif untuk flavonoid, dan Quercetin terdeteksi pada puncak pertama (2.329 menit) dengan konsentrasi 66.903 ppm. Analisis in silico menggunakan molecular docking memprediksi interaksi antara Quercetin dan jalur oksidatif terkait MDA. Quercetin bertindak sebagai inhibitor kompetitif terhadap enzim NADPH oksidase (NO), menunjukkan afinitas pengikatan sebesar –8,4 kkal/mol dan RMSD sebesar 1,904. Temuan ini menunjukkan bahwa Quercetin dalam teh Syzygium oleana dapat secara efektif mengurangi kadar MDA dengan menghambat aktivitas NO dan dengan demikian membantu mencegah kerusakan oksidatif pada perokok aktif

 

 

Smoking contributes to an increase in free radical levels, leading to oxidative stress in the body, which is marked by elevated levels of malondialdehyde (MDA). Malondialdehyde is a final product of lipid peroxidation that reflects DNA alterations, oxidized base compounds, and is strongly linked to the degenerative diseases. This study aims to evaluate the potential of Syzygium oleana in reducing MDA levels. The research includes antioxidant testing, phytochemical screening and identification, also in silico analysis. Antioxidant testing in Syzygium oleana tea showed an IC₅₀ value of 69 ppm, indicating strong antioxidant activity. Phytochemical analysis in the tea revealed a positive result for flavonoids, and Quercetin was detected at the first peak (2.329 minutes) with a concentration of 66.903 ppm. In silico analysis using molecular docking predicted the interaction between Quercetin and MDA-related oxidative pathways. Quercetin acts as a competitive inhibitor to the NADPH oxidase (NO) enzyme, showing a binding affinity of –8.4 kcal/mol and RMSD of 1.904. These findings suggest that Quercetin in Syzygium oleana tea may effectively reduce MDA levels by inhibiting NO activity and thus help prevent oxidative damage in the active smokers.
