Abstract

Tanaman buah ajaib (Synsepalum dulcificum) merupakan spesies dari famili Sapotaceae yang memiliki habitus semak dan berdaun tunggal yang berpotensi sebagai tanaman obat. Penelitian ini bertujuan untuk mengkarakterisasi anatomi dan histokimia daun buah ajaib serta menganalisis potensinya sebagai tanaman obat dengan menggunakan metode parafin, metode paradermal, dan histokimia. Struktur anatomi yang diamati mencakup ibu tulang daun, intercostal, tepi daun, dan tangkai daun. Hasil kajian anatomi tepi daun tersusun atas epidermis, jaringan palisade dan jaringan bunga karang. Ibu tulang daun memiliki berkas pengangkut berbentuk semi-sirkular. Tangkai daun berbentuk sirkular dengan berkas pengangkut flat convex. Stomata hipostomatik dengan tipe anisositik dengan kerapatan 31,13 ± 2,83/mm2. Trikoma non glandular dan trikoma glandular ditemukan pada permukaan adaksial dengan kerapatan trikoma non glandular 1,01 ± 0,22/mm2 dan kerapatan trikoma glandular 12,61 ± 0,00/mm2. Hasil uji histokimia menunjukkan bahwa daun buah ajaib positif mengandung senyawa metabolit dengan kepekatan warna yang bervariasi pada setiap jaringan yang diuji. Keberadaan senyawa tanin banyak ditemukan di semua bagian ibu tulang daun, intercostal, tepi daun dan tangkai daun. Senyawa flavonoid banyak ditemukan di tangkai daun dan tepi daun, senyawa terpenoid banyak ditemukan di ibu tulang daun dan tepi daun, dan senyawa alkaloid banyak ditemukan di tepi daun.

 

The miracle fruit (Synsepalum dulcificum) is a species from the Sapotaceae family with a shrub habit and simple leaves, which has potential as medicinal plant. This study aims to characterize the anatomy and histochemistry of the miracle fruit leaves and analyze their potential as a medicinal plant using paraffin, paradermal, and histochemical methods. The anatomical structures observed included the midrib, intercostal, leaf margin, and petiole. The anatomical study revealed that the leaf margin consists of the epidermis, palisade tissue, and spongy tissue. The midrib has a semi-circular vascular bundle, while the petiole is circular with a flat convex vascular bundle. The stomata are hypostomatic with an anisocytic type, having a density of 31.13 ± 2.83/mm2. Non-glandular and glandular trichomes were found on the adaxial surface, with non-glandular trichomes density at 1.01 ± 0.22/mm2 and glandular trichomes density at 12.61 ± 0.00/mm2. Histochemical test confirmed the presence of metabolic compounds with varying color intensities in the tested tissues. Tannins were widely found in all parts, including the midrib, intercostal, leaf margin, and petiole. Flavonoid were abundant in the petiole and leaf margins, terpenoids were mostly found in the midrib and leaf margin, while alkaloids were predominantly found in the leaf margin
